合理贮藏花粉并鉴定其生活力可以确保杂交育种获得较多群体数量的后代(Parton et al.,2002;王文莉 等,2009)。本研究中,川鄂连蕊茶的新鲜花粉萌发率可达 90%以上,说明本种的花粉生活力较强,且本试验中花粉采集时期及处理方法都较为适宜。本研究结果表明,川鄂连蕊茶花粉 4 ℃贮藏 60 d 时萌发率略有降低,萌发率仍大于 80%,这与平阴玫瑰较为相似(王文莉 等,2009)。但是在随后的 30 d,花粉萌发率大幅降低至 50%左右。虽然仍可满足杂交
授粉的需要,但得到杂交子代的群体数量将会有所减少。一般山茶品种的花期多处于 2 月至 4 月下旬,而川鄂连蕊茶的盛花期在 2 月至 3 月下旬,如果以本种作父本开展与山茶之间的杂交育种,花粉贮藏 30 ~ 60 d 足以解决亲本间花期不遇的问题。同时,作者在研究中还发现,贮藏的川鄂连蕊
茶花粉直接进行培养不能萌发,而经过 5 ~ 6 h 的室温吸湿处理后即可正常萌发,说明经过硅胶干燥处理后贮藏,川鄂连蕊茶花粉极易失水,如果不经过吸湿而直接播种于培养基上极易造成花粉粒外壁陡然吸水膨胀而造成损伤,从而导致花粉活力丧失。而经过吸湿处理后,花粉经过一个环境湿度过渡阶段,再进行离体培养时较易适应培养环境而正常萌发。
